Echinacea is frequently consumed in the hopes of enhancing immunity. Mixed findings have emerged from studies.

All parts of the Echinacea plant, including the base, leaves, flowerheads, and seeds, are used to create medicinal extracts. These are frequently employed to stop, treat, and shorten the duration of upper respiratory infections such colds and the flu (review in: [7]). Additionally, echinacea is used to treat infections and skin diseases such candidiasis (including ethnobotanical accounts of usage for thrush [1]), inflammations brought on by a range of bacteria, viruses, and other microorganisms linked to wound infections, as well as to reduce swelling and pain [812].
Several chemical components are thought to play a significant role in the therapeutic actions of echinacea. Alkamides, alkaloids, caffeic acid derivatives, flavonoids, essential oils, polyacetylenes, and polysaccharides are some of them [1317]. However, compared to other Echinacea ingredients like cichoric acid and caffeic acid, the alkamides, the main lipophilic molecules found in large concentrations in the roots of Echinacea, have a far better bioavailability [1820]. As a result, they might be to blame for the pharmacological effects of ethanolic extracts [21].
Echinacea has immunomodulatory properties that have been observed in in vivo and in vitro pharmacological studies [12,22], which may explain why it is effective in treating and preventing colds and the flu [7]. Echinacea’s antifungal and topical mechanisms of action, however, have not been thoroughly studied. Studying potential antifungal mechanisms of Echinacea extracts was our goal. Eight distinct ethanolic extracts of E. purpurea and E. angustifolia—a total of 16 treatments—were examined for potential phototoxicity and light-independent (dark) toxicity to Saccharomyces cerevisiae. We used a collection of 4,600 live S. cerevisiae gene deletion mutants with defined deletions of known or hypothetical genes to investigate the potential mechanism of action of echinacea. In order to assess the chemical-genetic interactions and pinpoint the pathways impacted by exposure to Echinacea extracts, the mutants were grown with and without each treatment.
The aforementioned strategy was chosen for three reasons. Based on the conventional use described above, we first looked into the mode of antifungal activity in extracts of all E. purpurea and E. angustifolia sections. Second, although the precise functions of the numerous lipophilic components in these extracts are mostly unknown, dark and near-UV light-enhanced antifungal activity of ethanolic extracts from these plants is well-established [11,23]. Therefore, we searched for common activities across various ethanol extracts of echinacea that had and hadn’t been activated by UV radiation. Last but not least, crude extracts’ chemical-genomic profiles can closely mirror those of their active, pure constituent substances [24]. The identification of the active compound(s) in complex combinations may therefore be guided by the mode of action determined by crude extracts.
The number of supersensitive yeast mutants with abnormalities in cell wall-associated processes was a significant pattern in our Echinacea gene deletion array (GDA) data sets, indicating that Echinacea extracts disrupt fungal cell wall biosynthesis and functionality. Then, we investigated whether fungi were more susceptible to sonication-based cell wall disintegration when cultivated in the presence of sub-inhibitory doses of Echinacea.
