Riffell’s team confirmed prior findings from field stations in Washington state’s Okanogan-Wenatchee National Forest that although this species is pollinated locally by mosquitoes, it is not pollinated by its close cousins that grow in the same habitat. The mosquitoes would still settle on the bagged flowers and try to eat through the canvas when researchers covered the blooms with bags, depriving the insects of a visual cue for the flower. Clearly, the mosquitoes were drawn to the orchid aroma. Riffell’s team looked at the various compounds that make up the aroma of the blunt-leaf orchid to understand why.
According to Riffell, “we frequently describe’perfume’ as if it were one entity, like the scent of a flower or a person. “The perfume of a rose actually comprises of more than 300 different chemicals, and mosquitoes can identify the different kinds of chemicals that make up a scent.”
The blunt-leaf orchid’s perfume, according to Riffell, is grassy or musky, in contrast to the sweeter scent of its cousins. The team identified several compounds in the fragrances of the Platanthera species using gas chromatography and mass spectroscopy. The blunt-leaf orchid smelled differently from its cousins in that it had higher concentrations of nonanal and lower concentrations of lilac aldehyde.
The electrical activity of mosquito antennae, which sense smells, was also captured by Riffell’s team. The native mosquitoes that inhabit the habitat of the blunt-leaf orchid had their antennae activated by both nonanal and lilac aldehyde. The Anopheles stephensi mosquito, which transmits malaria, and the Aedes aegypti mosquito, which transmits dengue, yellow fever, Zika, and other diseases, were among the mosquitoes from other locations whose antennae were likewise stimulated by these substances.
A mixture of nonanal and lilac aldehyde combined in the same proportion as found in blunt-leaf flowers was favoured by both native and non-native mosquitoes, according to studies on their behavior. Mosquitoes lost interest if the researchers left lilac aldehyde out of the formula. Mosquitoes were either unaffected by the aroma or turned off by it if they added more lilac aldehyde, at concentrations similar to those reported in near cousins of the blunt-leaf orchid.
They also looked into the brains of Aedes increpitus mosquitoes, which resemble blunt-leaf orchids, and a genetically altered strain of Aedes aegypti previously created by Riffell and co-author Omar Akbari, an associate professor at UC San Diego, using methods established in Riffell’s lab. They captured images of calcium ions, which are indicators of neurons that are actively firing, in the mosquito brain’s antenna lobe, which is responsible for processing data from the antennae.
According to these brain imaging studies, nonanal and lilac aldehyde stimulate various areas of the antenna lobe, and when triggered, they even compete with one another: The area that reacts to nonanal can inhibit activity in the area that reacts to lilac aldehyde, and the opposite is also true. The concentrations of nonanal and lilac aldehyde in the flower’s original aroma are likely what determines whether this “cross talk” renders a flower appealing to or repulsive to the mosquito. In contrast to closely similar species, blunt-leaf orchids have a ratio that repels mosquitoes, according to Riffell.

The ratio of certain molecules in human scent—which is quite complex—probably plays a key role in whether mosquitoes are attracted to or repelled by a person. We are aware that some people receive a little bit more than others, and perhaps the reason for this lies in the ratio.”
The group also found that DEET and lilac aldehyde both stimulate the same area of the antenna lobe. Although further research is needed to confirm this, that region may process “repressive” odors, according to Riffell. It’s too early to say whether lilac aldehyde will ever work as a mosquito repellent. But there is an added benefit if it is.
